Metabolicregulation of immune dysfunction during the progression of NASH toNASH-HCC

Description:

Thelack of effective treatments for non-alcoholic steatohepatitis(NASH) and NASH-associated hepatocellular carcinoma (NASH-HCC)underscores the need to understand the transition from NASH toNASH-HCC. Identifying why some NASH patients develop HCC whileothers do not is a significant challenge, with growing evidenceindicating that immune dysregulation plays a crucial role in thisprogression. The interstitial space, where cells receive nutrientsand signals, significantly impacts immune function, suggesting thatunderstanding the liver"s metabolic environment could reveal howimmune cells behave during NASH progression to HCC. By profilinginterstitial fluid metabolism and single-cell transcriptomics, theteam aims to understand the metabolic regulation of the immunelandscape in NASH to NASH-HCC progression. Using a high-fat,high-fructose, and high-cholesterol dietary mouse model thatmirrors human progression, the team will study the metabolic andimmune changes leading to NASH-HCC. In addition, the team'spreliminary findings show immune alterations and reduced levels ofthe bile acid hyodeoxycholic acid (HDCA) during NASH-HCCtransition. The identified candidate will also test the potentialof HDCA in preventing NASH to HCC transition, and further exploreHDCA"s effects on immune cells.

Jobresponsibilities:
  • Conduct in vitrowet lab experiments (e.g. cell culture, molecular biology andassays, microscopy etc)
  • Conduct in vivo animalexperiments (e.g. diet/treatment regimes, body weight &composition analyses, metabolic phenotyping, blood samplingetc)
  • Conduct end-point animal experiments(e.g. terminal blood collection, tissue collection and samplepreparation, whole tissue/organ imaging, serum measurementsetc)
  • Analyze collected data and maintainaccurate monitoring and reporting of experimental set-up, resultsand research findings
  • Interact and collaboratewith and assist other researchers and collaborators on relatedprojects
  • Communicate regularly with supervisorand join discussions with Principal Investigator and rest of thelab to discuss scientific progress and direction
  • Assist in the preparation of grant applications and grantprocess reports
  • Present findings both orallyand in written form via presentations and publication inpeer-reviewed journal
  • Assist in thepreparation of figures and manuscripts on the research projects forpublication in peer-reviewed journals
  • Contribute to other program-related projects andorganizations and with the general scientific community acrossA*STAR
