**Project Title**:Marine Organisms as Sources of Specialized Metabolites to Disrupt Bacterial Communication: Isolation and Identification of Quorum Sensing Inhibitors

**Project Introduction**: The emergence and the spread of multidrug-resistant bacteria constitute a major public health issue. Among incriminated Gram-negative bacteria, Pseudomonas aeruginosa has been designated by the WHO as a critical priority threat. P. aeruginosa has the capacity to form biofilms, reinforcing its virulence and intrinsic drug resistance. The regulation of biofilm and virulence factor production of this microorganism is controlled by a specific bacterial communication system called Quorum Sensing (QS). The development of antivirulence agents targeting QS that could attenuate P. aeruginosa pathogenicity without affecting its growth is therefore an attractive new therapeutic strategy. The current research proposal aims to identify unique specialized metabolites as QS inhibitors (QSIs) from largely underexplored local marine fauna and flora, specifically marine sponges, octocorals and cyanobacteria found at southern islands of Singapore. The proposal has four main objectives: (A) collection and screening of selected marine taxonomic groups for QS inhibitory activities using P. aeruginosa reporter strains, (B) perform MS-based metabolomic analysis of bioactive fractions for compound dereplication and detection of new/novel metabolites, (C) purification and structural determination of bioactive molecules using chromatographic and spectroscopic methods and (D) evaluation of compounds in attenuating virulence factors production and biofilm formation in P. aeruginosa with collaborators at SCELSE, NTU.

**Requirements**:

- Possess a BSc degree in Chemistry/Biology-related fields
- Be familiar with experimental techniques pertaining to organic chemistry and/or microbiology
- Ability to work independently
- Strong written and verbal communication skills
- Well-rounded and mature character
- Good interpersonal and organizational skills

**Responsibilities**:

- Assist in field collection of marine samples
- Carry out extractions of marine samples
- Carry out semisynthesis of organic molecules and microbiology-related bioassays
- Perform data analysis
- Other tasks as assigned by the Principal Investigator
