The Ramadan Group-Singapore is a newly established researchgroup located at LKCMedicine/NTU. Led by Prof Kristijan Ramadan,who has previously headed a well-established research group ingenome stability and cancer research at the University of Oxford,UK (The Ramadan Group-Oxford).

The RamadanGroup-Singapore is actively seeking an enthusiastic, hardworking,motivated, creative, and highly competent research fellow(post-doc) interested in addressing key biological questions inDNA-protein crosslink proteolysis repair. The Ramadan lab hasrecently discovered DNA-protein crosslink proteolysis repair as aspecialised DNA repair pathway for the repair of covalentlyattached proteins to DNA, known as DNA-protein crosslinks. Incollaboration with human geneticists, the laboratory has alsoidentified Ruijs-Aalfs Syndrome, the first human disease associatedwith biallelic and monogenic mutations in the SPRTN gene/protein,the key enzyme in DNA-protein crosslink proteolysis repair. Thisnovel DNA repair pathway has emerged as an essential mechanism forthe protection against accelerated aging and liver cancer, and theupregulation of this repair mechanism is likely linked to cancerpatient resistance to variouschemotherapies.

The post holder will beresponsible for thefollowing:
  • Perform aCRISPR/Cas9 screen in several mammalian celllines.
  • Validate the identified hits for theirrole in DNA repair.
  • Execute follow-upexperiments to generate a sufficient amount of results for ascientific publication.
  • Manage own academicresearch and administrative activities. This involves small-scaleproject management to coordinate multiple aspects of work to meetdeadlines.
  • Adapt existing and develop newscientific techniques (specifically CRISPR/Cas9 screen, cellsurvival, laser micro-irradiation, and DNA combing) and writedetailed experimental protocols.
  • Testhypotheses and analyse scientific data from various sources,reviewing and refining working hypotheses asappropriate.
  • Contribute ideas for new researchprojects related to DNA-protein crosslink (DPC) repair and genomestability.
  • Develop ideas for generatingresearch income and present detailed research proposals to seniorresearchers.
  • Collaborate in the preparation ofscientific reports and journal articles and occasionally presentpapers and posters.
  • Use specialist scientificequipment (e.g., DNA combing, Live Cell Imaging and Analysis) in alaboratory environment.
  • Act as a source ofinformation and advice to other members of the group on scientificprotocols and experimentaltechniques.
  • Represent the research group atexternal meetings/seminars, either with other members of the groupor alone.
  • Carry out collaborative projects withcolleagues in partner institutions and researchgroups.
  • Provide guidance to less experiencedmembers of the research group, including postdocs, researchassistants, technicians, and PhD and projectstudents.
  • Assist the Principal Investigator(PI/Prof Ramadan) in further grant writing andsubmission.
  • Assist in managing daily labduties.
  • Securely store and manage obtainedscientificdata.


KeyCompetencies:

  • Knowledgeof basic biomedical laboratory techniques, especially in the fieldof genome stability, ubiquitin system and geneediting
  • Experience: At least one first-authorpublication related to genome stability and/or cancer field in areputable international scientific journal, in addition to severalother original scientific publications in thefield.
  • Skills: Cell culture, molecular cloning,gene editing (CRISPR/Cas9), basic biochemical and cell biologicaltechniques (e.g., protein purification, Western blotting,immunofluorescent microscopy), willingness to learn newtechnologies.
  • Competence: highly skilled invarious basic wet laboratory tehniques and capable of successfullydelivering research goals in the highly competitive academicenviroment
  • Essential Qualifications: PhD inbiomedical research or nearcompletion
